چکیده
objective:glioblastoma is an invasive tumor of the central nervous system. epigenetic therapy of cancer is potentially very useful in reversing some of cancer defects due to reversibility of epigenetic alterations. meg3 is a tumor suppressor long non-coding rna (lncrna) that expresses in the majority of normal tissues. methylation of the meg3 promoter region elicits a decrease in its expression in glioblastoma cells. bioactive nutrients including curcumin offer great potential in altering dna methylation status. herein, we aim to investigate the epigenetic-based role of dendrosomal-curcumin (dnc) in upregulation of meg3 expression in glioblastoma cells. methods: we evaluated dnc entrance to u87mg cells with the use of the fluorescent characteristics of curcumin. next we performed the mtt assay to evaluate dnc and dendrosome effects on cell viability. the ability of dnc to boost expression of meg3 in dna methylation regulation was accomplished by a study of the relative expressions of meg3 and dna methylation regulator enzymes, dna methyltransferases (dnmt1, dnmt3a and 3b) using semi-quantitative and quantitative pcr. results: we observed the entrance of dnc into u87mg cells. dnc significantly caused u87mg cell death in a time and dose-dependent manner. however dendrosome did not show any toxic effect on this cell line. data acquired from gene expression assays determined that dnc upregulated meg3 expression (p

Glioblastoma is an invasive tumor of the central nervous system. Tumor recurrence resulting from ineffective current treatments, mainly due to the blood-brain barrier, highlights the need for innovative therapeutic alternatives. The recent availability of nanotechnology represents a novel targeted strategy in cancer therapy. Natural products have received considerable attention for cancer thera...
متن کاملEvaluating the clinical importance of long-non coding RNA MALAT1 expression in breast cancer
Background: Breast cancer is one of the major causes of illness and mortality among women. Long non-coding RNAs (LncRNAs) have important role in tumor development and progression. Metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) is a lncRNA that deregulates in several cancers, however, its value in the diagnosis of breast cancer is unclear. This study was conducted to investigate...
متن کاملDysregulated Expression of Long Non-Coding RNA MINCR and EZH2 in Colorectal Cancer
Background: As critical regulators, lncRNAs have attracted attention from researchers for diagnostic, prognostic, and therapeutic purposes in human carcinogenesis via interfering with mRNAs such as EZH2. Nevertheless, the potent roles and molecular mechanisms of these RNAs in CRC are not clearly known. Methods: In this study, the tissue expressions of lncRNA MINCR and EZH2 mRNA between colorect...
متن کاملmicroRNA-22 can regulate expression of the long non-coding RNA MEG3 in acute myeloid leukemia
AIM Acute myeloid leukemia (AML) is the most common blood tumor with poor prognosis. At present, the research found that the pathogenesis of AML is related to many factors, such as recurrent somatic mutations and gene expression and epigenetic changes, however, the molecular mechanism of AML is still unclear. Long non-coding RNA MEG3 is a newly found tumor suppressor and plays a very important ...
متن کاملSOX2OT, a long non-coding RNA involved in autophagy regulation
Summary: SOX2 overlapping transcript (SOX2OT) is a long non-coding RNA associated with cancer pathogenesis. It contributes to a variety of cellular functions and recent evidence propounds its association with autophagy process. It has been showed that SOX2OT can regulate the expression of different autophagy associated factors in human cells with different mechanisms, however more remains to ...
متن کاملP87: The Role of the Long Non-Coding RNA Sequences (LncRNAs) in Neurological Disorders
Precise interpretation of the transcriptome sequences in the several species showed that the major part of genome has been transcribed; however, just a few amounts of the transcription sequences have open-reading frames which are conversed during the evolution. So, it is unlikely that many of the transcribed sequences code the proteins. Among the all human non-coding transcripts, at least 10000...
